Photochemical internalization

چکیده
Introduction/aims Photochemical internalization (PCI) is a novel technology facilitates the delivery of macromolecules into cytoplasm. The initial mechanism and practical application was described by Berg et al. in 1999. This, first in human trial, is an open, phase I dose escalating study to evaluate the safety and tolerance of the photosensitizer (amphinex) that is used to initiate the photochemical internalization process with bleomycin as the chemotherapeutic agent. We present our preliminary report following the management of 11 patients with head and neck tumours.

متن کاملPhotodynamic therapy vs. photochemical internalization: the surgical margin
Controlling tumour margins in head and neck surgery is of the utmost importance in preventing loco-regional spread and distant metastasis, which will ultimately lead to a significant reduction in morbidity and mortality. We comment on the surgical margins in photodynamic therapy and photochemical internalization.

In many drug delivery strategies, an inefficient transfer of macromolecules such as proteins and nucleic acids to the cytosol often occurs because of their endosomal entrapment. One of the methods to overcome this problem is photochemical internalization, which is achieved using a photosensitizer and light to facilitate the endosomal escape of the macromolecule. In this study, we examined the m...

Because peptide nucleic acids (PNAs) are poorly taken up by mammalian cells, strategies need to be developed for their intracellular delivery. In the present study, we demonstrated the possibility to efficiently release a naked PNA targeting the catalytic component of human telomerase reverse transcriptase (hTERT-PNA) into the cytoplasm of DU145 prostate cancer cells through the photochemical i...
متن کاملComparing the Effects of Light- or Sonic-Activated Drug Delivery: Photochemical/Sonochemical Internalization.
Photochemical internalization (PCI) is a technique that uses the photochemical properties of photodynamic therapy (PDT) for the enhanced delivery of endolysosomal-trapped macromolecules into the cell cytoplasm. The released agent can therefore exert its full biological activity, in contrast to being degraded by lysosomal hydrolases. Activation of photosensitizers via ultrasound (US), called son...
